Demo 3 (N-Glycan from human IgG)

To test the Peak Detection Wizards validity, N-glycan mixture extracted from human IgG was used as an example of complex biological sample.

This mixture have been dissolved in MeOH/H2O 1/1(v/v) and submitted to nanoESI Q-Tof MS analysis in the positive (Demo 3(+) mode MS) ion mode

A library of N-glycans previously discovered in IgG was used as a reference data for system evaluation (Figure 3).

Due to structural isomericity, 31 N-glycan IgG structures presented on Figure3 corresponds to 21 unique glycan compositions. Out of them, 16 compositions (highlighted on Figure 3) were unambiguously assigned (Structure Nr: 01-13, 16-17, 22-28, 30).

To demonstrate MS/MS interpretation approach the precursor ions at m/z 1647 related to three potential structural candidates (08, 09 and 10 on Figure 3) have been fragmented and submitted for de novo N-glycan automated MS/MS interpretation (Figure 4).
